few cells microarrays

Postby denans » Jun 26 2008 4:52 pm

Hi all,

I'm new in microarray and even in molecular biology. I have to do some microarrays experiment (all chicken genome) on electroporated cells from chicken embryos. I got less than 100 electroporated cells by embryo. I use the affymetrix protocol for mRNA amplifications but I don't get enough mRNA to hybridize the chips with only one embryo. With this protocol I need around 25 embryos to get enough mRNA but this huge amount of embryos introduce a bias in my experiment. I know that some teams do microarrays on single cells. Does someone know a good protocol for mRNA amplification from few cells?

Thanks.
