On-chip PCR | Gene Frames etc

Postby Charles23 » May 05 2009 7:47 am

We try to establish solid phase PCR in our laboratory.

Our first setup comprises spotted microarrays on slides (GeneWave, Corning,...) that bordered and seal with GeneFrames (peqlab).

Unfortunately, these frames are not leak-tight and after initial activation of the polymerase (7' @ 95°C) all the PCR mix is evaporated.
Of course we used the manufacturer's protocol and tried some variations.

Has anyone of you experience with setting up slides for on-chip PCR?
Which components do you use?

Thank you in advance for feedback,
