IL-2 conversion to Units

Postby Whelans2 » Jun 08 2016 6:58 pm

Hi there,

I have 25ug/ml from R&D (202-IL) I need 100 U/ml to treat 40 wells that current have 200ul of cells in each.I think the activity is 2.1 x 10^4 IU/ug.I have stock 25ug am I right that this is 5250000U. I need to do 1:525 dilution and 1:100 to yield 100U/ml? So I need 50ul of IL-2 and 4950ul media?

Your stock is 25 ug/ml or 52.5 x 10^4 IU/ml (525000 IU/ml). So after 1:525 dilution, it will be 1000 IU/ml. Then the second dilution is 1:10 (not 1:100) to give 100 IU/ml.

What is the final concentration in the cell culture?

I meant the final concentration of IL-2 in the cell culture medium.
