qPCR reaction with probe self-dimer

Hello everyone,

I am trying to optimize a qPCR reaction. Unluckily, I realize that my probe form a self-dimer structure which impair the signal of amplification. How can I improve my qPCR reaction? I hope that someone can help me. :cry: :cry: :cry: :cry:

How did you discover the self-dimer, are you sure that is impairing the reaction?

What type of fluorophore and quencher are you using?

Redesign your probe if possible - if you can't change the sequence then try ordering from a different company
