Labeling DNA FISH probes

Postby arturko » Nov 29 2006 4:36 pm

I looking for complite protocols starting from BAC gene clones DNA isolation , labeling probes , applaying probes on formalin fixed tissues.

Anybody know about:

are BAC clones form Sanger Institute UK completely free for non commercial Institutes?

BAC clones...

Postby samward » Feb 22 2007 3:26 pm

We buy our BAC clones from BAC/PAC resources online.

We've had a lot of success growing up the clones and extracting the DNA using Qiagens Plasmid extraction kit. We then label the probe with SpectrumRed/Spectrum Green (Vysis) using their nick translation kit - 10┬Ál enzyme and an 8 hr incubation.

Hope this helps

We get our BAC clones from:

http://bacpac.chori.org/

They have good links to libraries as well. Delivery is really quick and we've had 100% success rate from their clones.

Re: Labeling DNA FISH probes

hi i would like to ask if 3 prime end labelling of FISH probes would be an advantage over 5 prime end labelling? I requested for a 3 prime end alexa 488 labelling of a FISH probe but i just learned they do only offer the 3 prime end alexa 488 labelling.

thank you.
