My student has just found out that vectashield quenches Alexa 546 (red) fluorescence. The company confirms this. What are alternative mounting media that people have had look luck with?

Hi!
We used 80% glycerol in phosphate buffer (PBS, pH 7.4) as a glycerol-based standard medium...
As anti-fading reagents, 0.1% p-phenylenediamine (PPD) or 2.5% 1,4-diazabicyclo-[2,2,2]-octane (DABCO) were added to the standard mounting medium.
We never used vectashield or any other commercial one...

I used that 0.1% p-phenylenediamine (PPD) compind for years on human, piglet and cultured cells, but just found that it makes huge nuclear autofluorescence into mouse tissue compare to VectaShield

Hmmm..... I have no problem with my Fluoroprobes 546 signal when I mounted my stained cells with Vectashield, but I found that Vectashield did not really protect my GFP signal from my transformed bacteria for more than 12 hours. Generally it only stabilized the GFP signal only for imaging. The GFP signal still bleached after imaging.
Confocal matrix from Micro-Tech-Lab works fine for me.

We used commercial mounting oil from Ab companies, such as DAKO.
Someone said woemn's nail oil is as good.
