Is it necessary a linker for knock-in GFP using CRISPR?

Postby Lauravuo » Apr 05 2016 6:13 am

Hi,

I am designing a strategy to knock in GFP at the N-terminus of one gene using CRISPR.

I read that for long insert the recommended length of the homology arms it is 800bp. Also I found that sometimes a liker sequence is knock in between the GFP  and the gene (for example GGTGGCGGC),

Do you think it is necessary?

Well i don't think it is necessary but if can get it, that would be better in my opinion. Using CRISPR would definitely made it better for sure.
