Cell density - confusion?

Postby livercell » Dec 13 2017 6:33 pm

Hi everyone,

I know it is an easy question for most of you but I want to check whether my calculation and reasoning is correct:

If I have a cell stock of 10 million cells in 8 mL, and I need to seed at 50,000 cells per well. This means, I have enough to seed 200 wells (so 3 - 96-well plates). I want to have 100 uL of 50,000 cells per well, i.e. 20 mL in total (100 uL per well x 200 wells = 20 mL) .

- This means I need to further add 12 mL of working medium to initial stock of 10 million cells in 8 mL to perform this experiment, i.e. I need to dilute a stock from 8 mL to 20 mL in order to obtain desired number of cells per well in a chosen volume.

Is this correct reasoning?

Thank you for your help.
